Studies on microsomal phospholipids that inhibit gulonolactone oxidase.

چکیده
Fractions of rat liver microsomal phospholipid that inhibit gulonolactone oxidase are described. The inhibition was shown to be complete and occurs rapidly after the reaction has proceeded at the control rate for 20 to 30 min. It could be totally reversed by the addition of tocopherol, Mn++, Co++, or ethylenediaminetetraacetate. The inhibitory activity of the phospholipid was lost by enzymic cleavage of the @-acyl fatty acid moiety or by hydrolysis of the bases associated with the phosphatidyl group. Diglycerides derived from the lipid retained significant inhibitory properties. The inhibition is apparently accompanied by alterations of unsaturated fatty acids resulting in formation of chromogenic substances that react with thiobarbituric acid. When the degree of unsaturation of the inhibitor phospholipid is reduced, both the inhibitory properties and formation of chromogens are proportionally decreased.

The enzyme system catalysing the conversion of D-glucuronolactone and L-gulonolactone into Lascorbic acid in animal tissues has been previously reported to be entirely located in the microsomal fractions ofratand goat-liver homogenates (Burns, Peyser & Moltz, 1956; Chatterjee, Ghosh, Ghosh & Guha, 1957a, b; 1958a, b). The kidney tissue has been found to be the site of this conversion in amphibi...

Solubilization and partial purification of the microsomal UDP-glucose sterol glucosyl transferase activity from maize coleoptiles by chromatography on DEAE-cellulose resulted in a highly delipidated (>95%) and inactive enzymic preparation. Addition of sterols revealed part of the activity and subsequent addition of phospholipids further increased the activity. Negatively charged phospholipids w...

Salinity is one of the most important abiotic stresses that limit crop growth and production. Salt stress influences plants in two ways: by affecting ion toxicity and increasing osmotic stress. Ion homeostasis, the excretion of Na+ and using antioxidant systems are the major strategies of salt tolerance in plants. Na+ and K+ transporters with enzymes that are involved in detoxification of react...

The importance of the deacylation-reacylation pathway for attaining the desired fatty acid composition in microsomal phospholipids has been well established. It is not clear, however, whether this mechanism is of equal importance in mitochondria. The absence of acyltransferase activity in mammalian heart mitochondria has been reported in a number of studies. In the present study we report the p...

Fe3+ was shown to be required for enzyme-catalyzed peroxidative chain-scission of polyunsaturated fatty acids of phospholipids in liver microsomal membranes. The K, value for iron in the reaction is different from that of nonenzymic processes which cause similar alterations of phospholipids. The products of both reactions include malondialdehyde which was shown to be the only substance formed w...
